Check sample identity before interpreting a figure
A measurement matrix and its metadata must use the same sample identifiers. Joining them by row position can silently assign the wrong labels after sorting or filtering. In a work sample, show the identifier check and what happens when a sample is missing or repeated.
Then inspect the design. A technically correct model cannot reconstruct information that was never observed. The following invented measurements illustrate that problem without using biological or patient records.
When the group and batch cannot be separated
Suppose group X was measured only in batch 1, with readings 9 and 11. Group Y was measured only in batch 2, with readings 19 and 21. The group means differ by ten units. But group membership and batch are perfectly tied together: the records do not identify how much of that difference comes from the group rather than the batch.
Subtracting ten from every batch-2 observation could remove a batch effect, a genuine group difference or both. The attractive corrected plot would not settle that question.
- In a different, deliberately balanced fictional design, batch 1 has group means X = 10 and Y = 12; batch 2 has X = 20 and Y = 22.
- Each batch now contains both groups. The within-batch difference is two units in each, while both groups shift by ten between batches.
- That comparison becomes possible because of the design, not because the second analyst chose a more sophisticated plotting tool.
Show both the analysis and the point where it must stop
Build a small reproducible notebook containing the two fictional designs, sample metadata and group summaries. State the difference between a descriptive pattern and an identifiable group comparison. Do not present the numbers as a biological discovery.
For a pipeline sample, add tests that reject duplicate sample IDs and identify metadata without a matching measurement. Preserve the inputs rather than quietly dropping unmatched rows. For an analysis sample, explain which extra observations would make the comparison more informative.
A useful application sentence is: 'Built a synthetic batch-effect example and identified when group and batch were inseparable from the available data.' It states a specific contribution without claiming expertise in an entire genomics workflow. Ask which data types and scientific questions the internship actually uses before choosing the final sample.
Before you apply
- Eligibility
- Genentech's FAQ includes several degree levels, at least one completed university year and graduates within two years for its internship program. Individual research roles can require narrower preparation.
- Timing
- Genentech describes summer postings from November through March. That is a recurring recruiting window, not a currently available bioinformatics assignment.
- Location
- Computational work is not automatically remote. Use the research group's stated worksite and ask what software and permitted data access the employer provides.
- Pay
- Genentech says internships are paid, with rates varying by degree and year. Its FAQ also describes housing or relocation support for interns covering their own housing costs; obtain the actual offer terms.
